脾细胞，从供体获取的脾脏，通过机械的方法或其他方法将脾组织捣碎制成游离的脾细胞保存于等渗盐水中即形成脾细胞悬液。脾细胞悬液的细胞总数用常规血细胞计数法算出，一个成人脾可制备550～12002亿个脾细胞。脾细胞存活率采用锥蓝染色法和相差显微镜两种方法对照测定。制定的脾组织悬液要求立刻应用，只作短期保存，保存期不宜超过6h。
